采用萃取法和反相HPLC技术分离蓝斑背肛海兔(Notarcus leachii cirrosus Stimpson,NLCS)卵内的产卵激素(egglaying hormone,ELH)及其分解产物.选用基质辅助激光解吸离子化飞行时间(Matrix-assisted laser deposition/ionization time-of-flight,MALDI)质谱技术研究NLCS卵内的ELH、ELH分解产物和酸性多肽(acidicpeptide,AP)二聚体的组成与结构特性.实验结果表明,NLCS卵不仅含有丰富的ELH及其分解产物,而且它的AP及其分解产物主要呈二聚体状态,推测这种二聚体结构对今后阐明ELH和AP的生理功能和调控机理起着重要的作用.
Aplysia is a kind of mollusk classing to be gastropod. The abundant drugs including hormones have been found in Aplysia egg. In order to understand the relation between Aplysia behavior and its hormones,the egg in Aplysia (Notarcus leachii cirrosus Stimpson) was selected to study the distribution and function of various hormones and peptides. The egg laying hormone (ELH) and its decomposed produces from the egg of NLCS were extracted by several organic solution and further separated by a combined off-line method of the extraction and the reversed phase- HPLC for analysis of molecular structures. The composition and structural characteristics of ELH,the decomposed produces of ELH,the dimer of acidic peptide were identified by MALDI-TOF mass spectrometry and Paws software. With reference to analytical methods as previously described, the experimental results show that the egg in NLCS not only contains abundance ELH and its decomposed produces,but also AP and its decomposed produces with dimeric molecular structure. In addition, we find that ELH is composed of AP and the decomposed peptldes of ELH,indicating that AP play an important role in signal transmission in egg, which the physical function of AP differs from that of ELH in egg. It is well known that ELH in egg see after carrying out the complex physiological function because it enables decomposing various peptides with different molecular structures.
