目的研究创伤后应激障碍(PTSD)样大鼠海马神经元溶酶体(LY)及线状溶酶体(NLY)的变化。方法利用大鼠PTSD-SPS模型,于SPS刺激后的6h,12h,1d,7d,14d取大脑海马,用溶酶体标记酶酸性磷酸酶(ACP)光电镜酶组织细胞化学技术,显示溶酶体,进行海马神经元溶酶体变化的观察。结果模型建立后的6h、12h细胞ACPase活性增强,溶酶体增多,1d时ACP活性更为显著,NLY尤为增加,7d、14d时ACPase活性减弱。结论PTSD样大鼠海马神经元1d时溶酶体表达最为显著,ACP酶活性最强,为了适应其变化NLY尤为增多。
Objective To study the lysosome (LY) and nematolysosomes (NLY) express in PTSD (posttraumatic stress disorder) animal model. Methods PTSD-SPS (single-prolonged stress) animal model was established, materials were taken at the 6 h, 12 h, 1 d, 7 d and 14 d after exposed to STXS. The changes of ACP expression of different SPS groups were detected by transmission electron microscopy and enzyme histochemistry. Results The lysosomes and activity of ACP were increased after 6 h and 12 h exposed to SPS. In addition, the activity of ACP and especially the NLY increased significantly 1 d after exposed to STXS, and the activity of ACP gradually decreased after 7 d and 14 d. Conclusion The expression of lysosomes is most significant 1 d after exposed to SPS and the activity of ACP reaches the top with the corresponding increase of the nematolysosomes (NLY) adapted to the former.
