采用酶法分别从雌、雄性皱纹盘鲍脏器中提取鲍鱼脏器粗多糖(AVP)。经Sephadex G-100凝胶过滤柱分离后,FAVP和MAVP各得到两个组分:FAVPⅠ,FAVPⅡ;MAVPⅠ,MAVPⅡ。它们清除羟自由基的EC50分别为1.38,0,99,1.51,1.19mg/mL,与对照Vc(EC50为1.23mg/mL)相比活性更强;其还原能力的A700,0.2分别为37.68,7.37,22.41,7.63mg/mL,与对照Vc(EC20为30.57μg/mL)相比弱很多;其络合能力则很弱。相关分析结果表明,鲍鱼脏器多糖具有一定的抗氧化能力。
The crude polysaccharides termed as AVP was extracted from female and male abalone viscera (Haliotis discus hannai Ino) by protease extraction. The female and male AVP (FAVE and MAVP) were fractionated by gel filtration chromatography with scphadex G -100, each giving two fractions containing polysaccharide conjugates termed FAVP Ⅰ , FAVP Ⅱ , MAVP Ⅰ , MAVP Ⅱ. The EC50 values of scavenging activity on hydroxyl radical of the four fractions were 1. 38, 0. 99, 1. 51, 1.19 mg/mL respectively, which were higher than that of positive control Vc(EC50: 1.23 mg/mL). The A700, 0.2 values of reducing power were 37.68, 7.37, 22.41,7.63 mg/mL respectively, which were lower than that of positive control Vc ( A700, 0.2 : 30.57 μg/mL). The chelating ability of the fractions was undetectable. Based on the correlation analysis, there were negative correlation between scavenging activity on hydroxyl radical and polysaccharide content. However, the exact relationship between the polysaccharide and protein content and antioxidant need further studies.
