目的 利用吗啡刺激小鼠小胶质细胞系BV-2细胞,研究NF-κB抑制剂PDTC对吗啡诱导的炎症因子表达的作用.方法 应用实时定量PCR检测 BV-2细胞炎症因子IL-1β、IL-6、TNF-α的表达.结果 吗啡处理BV-2细胞,可以导致IL-1β、IL-6、TNF-α的mRNA水平显著升高,而PDTC可显著抑制吗啡诱导的炎症因子mRNA水平的升高(P< 0.05),同时PDTC对基础状态下BV-2细胞IL-1β、IL-6、TNF-α的表达无显著性影响(P> 0.05).结论 PDTC显著抑制吗啡诱导的小胶质细胞活化,利用安全有效的药物来抑制NF-κB可能成为提升吗啡镇痛作用的新策略.
Objective We used morphine to stimulate murine microglia cell line BV-2 cells, to detect the effect of PDTC, the inhibitor of NF-KB, on the inflammatory factors expression induced by morphine. Methods The expression of inflammatory factors IL-1β,IL-6,TNF-α were measured by Quantitative Real-Time PCR. Results Morphine could significantly upregulate IL-1β, IL-6 and TNF-α mRNA levels; PDTC significantly suppressed the upregulatiou inflammatory factors induced by morphine (P 〈 0.05 ). Under the physiological status, there was no significant influence on expression of inflammatory factors in BV-2 cells(P 〉 0.05 ). Conclusion PDTC significant suppressed morphine induced microglial activation. Inhibiting NF-κB with safety and effective drugs can be a new strategy of improving morphine antinociceptive effect.
