目的:研究Bcl 10和IL-23在白念珠菌活化的树突状细胞内的表达变化规律及其意义。方法:采用热灭活白念珠菌体外刺激人单核源性树突状细胞,于第2、4、6和12小时以RT-PCR和Western blot检测Bcl 10的表达状况,同时采用RT-PCR和ELISA检测IL-23的表达。结果:Bcl 10的表达在刺激4小时后逐渐降低。白念珠菌刺激可以迅速诱导IL-23的表达,但IL-23的表达在4小时达到顶峰后即下调。结论:热灭活白念珠菌刺激的人单核源性树突状细胞Bcl 10表达降低,这可能是引起白念珠菌刺激4小时后IL-23表达降低的原因之一。
Objective:To investigate the expression patterns of Bcl 10 and IL-23 in heat-killed Candida albicans stimulated dendritic cells and it potential significance.Methods: Monocyte derived dendritic cells were stimulated with heat-killed Candida albicans.The expression of Bcl 10 at 2,4,6 and 12 hours after stimulation was determined by RT-PCR and Western blot.In addition,the expression of IL-23 was determined by RT-PCR and ELISA at the time point mentioned above.Results: Decreased Bcl 10 expression was observed after heat-killed Candida albicans stimulation.IL-23 expression was rapidly up-regulated by Candida albicans,reached peak at 4 hours after stimulation,and then decreased.Conclusion: Candida albicans stimulation down regulated the expression of Bcl 10 in dendritic cells,this may responsible for the decreased IL-23 expression after 4 hours of Candida albicans stimualation.
