目的:观察当归补血汤对体外造血微环境中小鼠肌源性干细胞分化的影响。方法:分离小鼠肌源性干细胞并培养鉴定。制备含有不同剂量当归补血汤的大鼠载药血清及对应剂量的骨髓基质细胞条件培养基。将肌源性干细胞随机分为8组:正常大鼠血清组、当归补血汤载药血清1~3组、正常大鼠血清条件培养基组、条件培养基1~3组,免疫组化法检测CD45的表达情况。结果:培养的肌源性干细胞呈desmin染色阳性;培养3天时载药血清3组、条件培养基两组及3组阳性细胞与正常大鼠血清组之间相比CD45表达量有显著性差异;随当归补血汤载药血清浓度增大CD45表达量增多,10倍剂量载药血清作用效果最佳,条件培养基也呈同样的变化趋势,经10倍剂量载药血清干预的条件培养基作用效果显著。结论:当归补血汤载药血清及含载药血清的条件培养基可促进肌源性干细胞向造血细胞方向分化。
Objective:To investigate the effect of Danggui Buxue Decoction on the differentiation of mouse muscle derived stem cells(MDSCs)in the different culture environment.Methods:MDSCs from new born mice were cultured and identified.Serum contained of Danggui Buxue Decoction and the marrow stroma cell conditioned medium were prepared.MDSCs were divided into 8 groups,and then MDSCs were cultured at different concentrations in the serum containing drug of Danggui Buxue Decoction and in the marrow stroma cell conditioned medium disposed by different concentrations in the serum containing drug.The CD45 protein of the MDSCs was examined by immunohistochemistry.Results:The desmin protein was positive in the cultured MDSCs.After the stimulation of various culture fluid,the results suggested MDSCs showed MDSCs of the Danggui Buxue Decoction containing serum group 3 and the marrow stroma cell conditioned medium group 2 and 3 could express more CD45 protein than cells of the normal rat serum control group.The Danggui Buxue Decoction containing serum could promote CD45 expression of the MDSCs in a dose-dependent manner,especially 10 times the dose of the Danggui Buxue Decoction containing serum.MDSCs of the conditioned medium groups were the same trend with MDSCs of the Danggui Buxue Decoction containing serum groups also.Conclusion:The Danggui Buxue Decoction containing serum and the marrow stroma cell conditioned medium disposed by the serum containing drug of Danggui Buxue could promote MDSCs to differentiate hematopoietic cells.
