嗅鞘细胞移植治疗自身免疫性脑脊髓炎的实验研究

目的研究嗅鞘细胞(OECs)的培养及其对自身免疫性脑脊髓炎(EAE)的治疗作用.方法取36只EAE模型大鼠,将其随机分为移植组18只,假手术组18只.用新生Wistar大鼠嗅球培养出OECs,移植组将OECs悬液注入EAE大鼠的侧脑室;假手术组按以上方法注入等量生理盐水.观察2组大鼠运动功能评分、组织病理学变化以及血管内皮生长因子(VEGF)表达情况.结果(1)OECs移植组运动功能评分从第1天的(6.8±0.7)分降至第20天的(1.1±0.3)分,显著优于假手术组,差异有统计学意义(P<0.05);(2)组织病理切片可见OECs移植组髓鞘修复明显优于假手术组;(3)OECs移植组VEGF的表达从第1天的(22.1±2.8)个/mm^2增至第10天的(300.6±5.1)个/mm^2,后逐渐降至第21天的(99.0±3.5)个/mm^2,在各时间点的表达均明显高于假手术组,差异有统计学意义(P<0.01).结论OECs移植可以促进EAE的髓鞘修复,改善运动功能,同时可使VEGF大量表达,诱导病灶周围新生血管的生成.

英文摘要:

Objective To investigate the culture of olfactory ensheathing cells (OECs) and the therapeutic effect of OECs transplantation on experimental allergic encephalomyelitis (EAE). Methods 36 male EAE model wistar rats were randomly allocated into OECs transplantation group and pseudo-operation group by halves. OECs suspension which was cultured with the olfactory bulb of newborn wistar rats was infused into the lateral cerebral ventriculus in OECs transplantation group. Simultaneously, the equal volume of saline was infused in pseudo-operation group. The movement score, histopathological changes and expression of VEGF were observed. Results ( 1 ) The movement score of OECs transplantation group, from 6.8 ± 0.7 on the first day dropping to 1.1± 0. 3 on the 20th day, was significantly higher than that in the pseudo-operation group (P 〈 0. 05 ) ; ( 2 ) Myelin reparation of OECs transplantation group was markedly better than that of pseudo-operation group in histopathological studies; ( 3 ) The expression of VEGF of OECs group, being (22.1 ± 2.8)/mm^2 on the first day, increased to (300.6 ± 5.1 )/mm^2 on the 10th day and then gradually dropped to (99.0 ±3.5)/mm^2 on the 21st day, which was statistically higher than that in the pseudo-operation group at each observation point (P 〈0.01 ). Conclusion OECs transplantation promotes myeline reparation in EAE model rats, improvement of movement function and expression of VEGF that induces blood vessels regeneration around lesions.
