目的:优选富集纯化石榴皮中总鞣质的大孔吸附树脂,确定工艺条件及参数。方法:采用10倍量70%丙酮水溶液50℃回流提取石榴皮中鞣质,并采用磷钼钨酸一干酪素比色法测定提取液中的总鞣质含量;通过静态与动态吸附和解吸性能的研究,对比分析了HP20、D-101、AB-8、HPD-450、HPD-400、NKA-9、HPD-500、HPD-826八种大孔吸附树脂对石榴皮中总鞣质的富集纯化效果,优化了HPD400型树脂富集纯化石榴皮中总鞣质的工艺条件。结果:8种大孔吸附树脂中,HPD-400型树脂对石榴皮中总鞣质有较好的吸附分离性能,其工艺条件为上样液质量浓度4.35H蜒·mL-1,吸附流速为2BV·h-1,上柱量为5BV(BV,树脂体积数),以70%的乙醇洗脱,用量为2BV,解吸率为93.94%,采用上述方法纯化后,产品中鞣质的纯度为60.49%。结论:HPD400型树脂对石榴皮中总鞣质纯化的综合性能较好,工艺的重复性和稳定性良好,为后续研究提供了基础。
OBJECTIVE To optimize the technological parameters for purified the total tannins in pomegranate peels by macroporous adsorbent resin. METHODS The tannins of pomegranate peels were extracted with 10 times the volume of 70% acetone by reflux extraction at 50℃. A modified tungsten molybdophosphate-casein colorimetric method was used to test the total tannin contents. The static and dynamic adsorption and desorption experiments were employed to investigate the effect of purification of tannins with eight kinds of macroporous adsorbent resins and the technological conditions of HPD-400 were opti mized. RESULTS Among the eight kinds of macroporous adsorhent resins, HPD-400 resin showed the best separating and pu rifled results. The content of sample solution was 4. 35 mg·mL-1 ,the velocity of flow was 2 BV·h -1 ,the maximum adsorbing capacity 5 BV. The best desorption rate was 93.94% with conditions of 2 BV of 70% ethanol as eluting solvent. By the method listed above the purity of tannins of pomegranate peels was measured to be 60. 49%. CONCLUSION HPD-400 resin is effec tive in the adsorption and purification of the tannins form pomegranate peels, with good repetition and stability.
