以3年生三七成熟的种子为材料,采用硅胶脱水(快速脱水)和自然风干(缓慢脱水)的方法处理种子,不定期测定种子超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性及种子萌发率.研究不同脱水速率对顽拗性三七种子生理生化特征及萌发率的影响,以期为三七种子选择合理的贮藏方法提供科学依据.研究结果表明,三七种子的初始含水量为67.3%,萌发率为11.11%;当含水量同样降至约60%时,快速脱水处理后的种子萌发率为5.56%,显著高于缓慢脱水处理后的种子萌发率(1.11%).随着脱水时间的延长,SOD、POD、CAT活性均呈现出逐渐下降的趋势,但在快速脱水状态下均基本高于缓慢脱水.脱水速率是影响三七种子脱水敏感性的重要因素之一;在一定的脱水程度上,缓慢脱水对三七种子的伤害更大.
The purpose of this study was to examine the effects of desiccation on the physiological and biochemical characteristics and the germination rate of recalcitrant Panax notoginseng seeds,and thus to provide the scientific basis to choose reasonable storage methods for Panax notoginseng seeds.Three-year-oldPanax notoginseng of mature seeds were treated by methods of silica gel desiccant (rapid desiccation) and naturally drying (slow desiccation).The result showed that the initial water content and germination rate of seeds was 67.3% and 11.11%,respectively.When the water content was lowered to about 60%,the germination rate was 5.56% in seeds treated by the rapid desiccation,which was higher than the germination rate of 1.11% in seeds treated by slow desiccation.The superoxide,peroxidase and catalase activity in seeds decreased slowly with the prolonged desiccation time,but they were higher in rapidly desiccated seeds than the in slowly desiccated seeds.Our results suggest that the desiccation rate was an important factor inducing the desiccation sensitivity of P.notoginseng seeds and to a certain extent the seeds of P.notoginseng were harmed greater by slow desiccation.
