目的探讨肝素酶抑制剂OGT2115对人口腔上皮癌KB细胞增殖及侵袭迁移能力的影响,以期为治疗肿瘤转移提供思路。方法 MTT法和Transwell实验分别检测不同浓度OGT2115(0.4、0.8、1.6、3.2、6.4μmol.L-1)对口腔癌KB细胞增殖以及侵袭迁移的影响;Transwell实验和划痕实验检测OGT2115(0.4μmol.L-1)联合低浓度(2μmol.L-1)阿霉素(adriamycin,ADM)后对细胞侵袭迁移的作用;ELISA法观察不同浓度OGT2115、OGT2115联合ADM对乙酰肝素酶(heparanase,Hpa)的影响。结果 MTT法结果显示,OGT2115对KB细胞的增殖具有一定的抑制作用,IC50为8.59μmol.L-1,抑制活性随着作用时间和浓度的增加而增加(P〈0.05)。OGT2115可抑制KB细胞的侵袭和迁移,OGT2115(0.4μmol.L-1)与ADM(2μmol.L-1)联用后可以明显增强ADM抗KB细胞增殖和侵袭迁移能力(P〈0.05)。结论肝素酶抑制剂OGT2115可以抑制KB细胞的侵袭和迁移,且可增强ADM抗KB细胞侵袭迁移的能力,其机制可能与抑制Hpa的活性有关。
Aim To investigate the inhibitory effect of heparanase inhibitor OGT2115 on the invasion and migration in KB cells,in order to provide a new target points for the treatment of cancer metastasis.Methods MTT assay was used to detect the cell viability by different concentrations of OGT2115(0.4,0.8,1.6,3.2,6.4 μmol·L-1) and OGT2115(0.4 μmol·L-1)combine with ADM(2 μmol·L-1)in KB.Transwell assay was applied to detect the invasion and migration by OGT2115 and ADM.Wound healing assay was employed to observe cell migration,and heparanase activity was detected by ELISA.Results MTT results showed that OGT2115 could inhibit the proliferation of KB;IC50 was 8.59 μmol·L-1;the inhibitory activity increased with the role of time and concentration(P<0.05).OGT2115 could inhibit the invasion and migration of KB;using OGT2115(0.4 μmol·L-1)significantly enhanced anti-invasion and anti-proliferation of ADM(P<0.05).Conclusion OGT2115 can inhibit the invasion and migration of KB,and OGT2115 enhances anti-invasion and anti-migration of ADM,which is closely related to heparanase.
