为利用高效液相色谱法(HPLC)—荧光检测器法测定催醒过程中小型猪血浆中的阿替美唑浓度方法,试验以Symmetry C18(4.6 mm×250 mm,5μm)为固定相,乙腈与去离子水(0.05 mol·L-1磷酸盐缓冲液)30 70(V/V)为流动相,流速0.8 mL·min-1等色谱条件进行测定。结果表明,该法回收率理想,最低检测限为100 pg·mL-1,阿替美唑分离效果良好。说明此法专属性强,准确度高,方法可靠,满足催醒过程中小型猪血浆中阿替美唑含量的测定及临床药代动力学研究要求。
To detect the concentration of atipamezole in plasma of small-sized pig by HPLC-FL,Symmetry C18(4.6 mm × 250 mm,5 μm) was used as stationary phase,acetonitrile and deionized water(0.05 mol.L-1 phosphate buffer) 30:70(V/V) as mobile phase,flowing rate was 0.8 mL.min-1.The result shows that the recovery rate is good,the limit detection is 100 pg.mL-1,the separation effect of atipamezole is good,demonstrating that the method is highly specific,accurate,as well as reliable,it comforts the detection of the concentration and the pharmacokinetic of atipamezole during recovery period.
