目的:探讨健脾解毒方通过抑制MALAT1介导的β-catenin信号通路抗大肠癌侵袭转移的作用机制。方法:利用慢病毒介导的RNA干扰技术沉默MALAT1基因,观察健脾解毒方对大肠癌细胞侵袭转移及MALAT1介导的β-catenin信号通路的调控作用。结果:健脾解毒方能够抑制大肠癌细胞的侵袭转移能力,直接下调MALAT1基因的表达,抑制大肠癌细胞胞核内β-catenin的积累及其下游靶基因MMP-7的表达;MALAT1基因能够增加β-catenin在细胞核内的积累及其下游靶基因MMP-7的表达。结论:MALAT1通过激活β-catenin信号通路并促进下游靶基因MMP-7的表达以达到促进大肠癌细胞侵袭转移的目的,而健脾解毒方能够通过直接抑制MALAT1的表达发挥抗大肠癌细胞侵袭转移的作用。
Objective: To investigate the mechanism of Jianpi Jiedu recipe(JPJDR) on the inhibition of the invasion and migration of colorectal cancer through MALAT1 mediated β-catenin signaling pathway. Methods: The lentivirus mediated RNA interfere technology was used to silence the expression of MALAT1. And then, how JPJDR to regulate the β-catenin signaling pathway mediated by MALAT1 and to influence on the invasion and migration of colorectal cancer were observed. Results: JPJDR inhibited the ability of invasive and migratory of colorectal cancer. JPJDR reduced the accumulation of β-catenin in the nuclear of colorectal cancer as well as the expression of MMP-7 gene, and importantly JPJDR directly down-regulated the expression of MALAT1 gene, which could increase the accumulation of β-catenin in the nuclear and promote the expression of β-catenin targeted MMP-7 gene. Conclusion: Jianpi Jiedu recipe could inhibit the invasion and migration of colorectal cancer through MALAT1 mediated β-catenin signaling pathway.
