猪瘟病毒(CSFV)和猪蓝耳病(猪繁殖与呼吸综合征)病毒(PRRSV)是引起严重的种猪繁殖障碍的病原,而且经常混合感染,及时准确诊断是防治的前提。根据已发表的猪瘟病毒(CSFV)E2基因的保守序列和猪繁殖与呼吸综合征病毒(PRRSV)ORF7基因的保守序列,设计合成了2对特异引物,分别扩增出大小为509 bp和372 bp的特异性基因片段,通过优化RT-PCR条件,分别对江苏不同地区送检的21份病死猪的血清、淋巴结、肺、肝、脾、心脏等组织进行单重RT-PCR检测。结果4份CSFV阳性,9份PRRSV阳性,其中3份CSFV和PRRSV同时为阳性。结果表明,CSFV和PRRSV单重RT-PCR诊断方法具有高度特异性,可用于临床诊断。
Classical swine fever virus(CSFV) and porcine reproductive and respiratory syndrome virus(PRRSV) are the major causes of some serious reproductive diseases,and co-infection between them is very common.Based on conserved sequences in E2 fragments of CSFV and those in ORF7 fragments of PRRSV,two pairs of specific primers are respectively designed,and the sizes of the amplified specific gene fragments are 509 bp and 372 bp.Twenty-one samples(including blood serum,lymphoid node,lung,liver,spleen and heart) from different farms in Jiangsu province are respectively detected by optimizing the conditions for RT-PCR,of which four are CSFV positive,nine are PRRSV positive,three are both CSFV and PRRSV positive,and the rest samples are negative.Therefore,the simplex RT-PCR of CSFV and PRRSV is a rapid and specific diagnostic method,which can be used in clinic diagnosis.
