目的分析羊栖菜多糖对S180荷瘤小鼠红细胞相关生化功能影响的研究。方法建立肿瘤动物模型,分高、中、低剂量腹腔给予羊栖菜多糖7d,采集红细胞,制备红细胞悬液,运用激光共聚焦扫描技术测定荷瘤小鼠红细胞内[Ca^2+]的变化;运用相关试剂盒测定红细胞膜表面唾液酸含量和Na^+,K^+.ATPase及Ca^2+,Mg^2+-ATPase活性的变化;用流式细胞仪分析红细胞膜电位水平的变化;采用高效毛细管电泳法测定红细胞电泳合淌度的变化。结果羊栖菜多糖能降低荷瘤小鼠红细胞内[Ca^2+],升高膜表面唾液酸的含量,增强膜表面Na^+,K^+-ATPase及Ca^2+,Mg^2+-ATPase的活性,提高红细胞膜电位水平,提高红细胞的电泳合淌度。结论羊栖菜多糖可调节或恢复S180荷瘤小鼠红细胞多种生理生化功能。
OBJECTIVE To study the effect of Sargassum fusiforme polysaccharides (SFPS) on biochemistry function of erythrocyte membrane in S180 mice. METHODS A tumor mouse model was developed, and the mice were treated with high, medium, and low doses of SFPS through abdominal injection for 7 d. Erythrocytes were collected and erythrocyte suspensions were prepared. Changes of [Ca^2+] in the erythrocytes of the tumor-bearing mice were measured using laser confocal scanning; changes of sialic acid and the acitivities of Na^+, K^+-ATPase and Ca^2+, Mg^2+-ATPase were meausred using relevant test kits; changes of erythrocyte membrane potential were analyzed using flow cytometry; and changes of the complex mobility of erythrocytes were measured using high performance capillary electrophoresis (HPCE). RESULTS SFPS decreased the intracelluar [Ca^2+] of erythrocytes in tumor-bearing mice, increased the content of sialic acid on surface of erythrocyte membrane, increased the acitivities of Na^+, K^+-ATPase and Ca^2+, Mg^2+-ATPase, and raised the level of erythrocyte membrane potential and the complex mobility of erythrocytes in elelctrophoresis. CONCLUSION Sargassum fusiforme polysaccharides (SFPS) can adjust or restore some biochemistry functions of erythrocyte membrane of S180 mice.
