目的探讨影响中期保存的供体角膜内皮细胞活性的相关因素。方法对保存液中的角膜内皮细胞进行培养和评分,并与供体年龄、供体保存与死亡时间间隔、供体采集与死亡间隔、是否曾在常温下长时间暴露等因素进行相关分析。结果细胞培养评分与供体年龄呈负相关,与供体保存时间呈负相关,与是否在常温下过夜呈负相关。结论供体的年龄、植片的保存时间及是否在常温下长时间暴露均是影响供体角膜内皮细胞活力和功能的关键因素。
Objective The aim of tissue engineering cornea is to develop substitutes for the replacement of wounded or diseased corneas.The residual donor corneas after penetrating keratoplasty are now serving as a main source for culture human corneal endothelial cells in vitro.This study was performed to investigate factors affecting vigor of the endothelial cells on donor cornea stored in corneal storage medium.Methods Corneal endothelial cells of 172 residual donor corneas from penetrating keratoplasty were cultured and digested.Cultured corneal endothelial cells were identified using S-100 protein.The cultured cells were classified into 0-6 scores based on the growth and passaged ability,and cornea with its score equal or more than 3 was considered to be valuable.Effects of donor age,exposure condition,storage time and time interval between enucleating and death on cell culture score were analyzed.Results The number of cornea with the score equal of more than 3 was 90.Donor age,exposure in room temperature and storage time showed negative correlation to cell culture score(r=-0.319,P〈0.01;r=-0.343,P〈0.01 and r=-0.229,P=-0.05,respectively).While time interval between enucleating and death had no correlation with statistical significance to cell culture score(r=0.092,P=0.229).The corneas score in≤40 years group was higher than that〉40 years(P〈0.01).The corneas score in room temperature exposure group was higher than that without room temperature(P〈0.01).The corneas score in≤360-hour preservation group was higher than that〉360-hour preservation group(P〈0.01).Conclusion Donor age,exposure in room temperature and storage time are important factors affecting the vigor of donor cornea stored in corneal storage medium.
