目的:探讨miRNA-196a-2的单核苷酸多态(single nucleotide polymorphisms,SNPs)位点rsll614913与胃癌易感性的关系。方法:采用病例一对照研究设计,研究对象为232例经组织学确诊的胃癌病例和250例按年龄、性别进行匹配的无肿瘤史的社区对照人群。采用探针法SNP基因分型技术检测miRNA-196a-2基因的C/T多态的基因型,计算rsll614913多态位点基因型在病例与对照中的分布情况,采用logistic回归计算校正相对危险度(oddsratio,OR)和95%置信区间(95%confidenceinterval,95%CI).评价基因型对胃癌易感性的影响。结果:miRNA-196a-2基因多态位点rsll614913CC、CT和1Tr基因型在胃癌人群中的分布频率为44.0%、47.0%和9.0%,与对照组(28.8%,54.4%和16.8%)相比差异有统计学意义(均P〈0.05);与CC基因型相比,携带1Tr或CT基因型的个体胃癌发病危险度降低(OR=0.369,95%CI:0.204~0.659;OR=0.564,95%CI:0.387~0.830);携带突变T碱基(CT/TT)的胃癌患者发生淋巴结转移的危险度降低(OR=0.618,95%CI:0.438~0.795;P=0.010)。讨论:miRNA-196a-2基因的C/T多态性与中国汉族人群胃癌的遗传易感性相关。
Objective:To determine the association between miR-196a-2 rs11614913 C/T polymorphisms and susceptibility of gastric cancer. Methods: In a case-control study, we investigated 232 patients with pathology-diagnosed gastric cancer and 250 age- and sex-matched healthy controls recruited from communities. This entailed genotyping of miR-196a-2 rs11614913 C/T polymorphisms by using Taqman SNP genotyping assay for calculation of the distribution of rs11614913 polymorphisms in eases and controls. Logistic regression analysis was employed to derive the adjusted odds ratio (OR) and 95 % confidence interval ( 95 % CI) for assessment of the association between the susceptibility of gastric cancer and rs11614913 genotypes. Results: The frequencies of genotypes CC, CT and TT were 44. 0% , 47. 0% and 9. 00/0 in patients with gastric cancer, which were significantly different from that in controls ( 28.8% , 54.4% and 16.8% , all P 〈 0.05 ). Compared with CC genotype ,homozygote and heterozygote variant forms( TT and CT)were characterized by significantly reduced risk of developing gastric cancer( OR = 0. 369,95% CI : 0. 204 - 0,659 ; OR = 0. 564,95% CI : 0. 387 - 0. 830 ). Patients with T allele mutants (CT/TT)had a reduced risk of having lymph node metastasis (OR = 0. 618,95% CI : 0. 438 - 0. 795 ). Conclusion : The miR-196a-2 rsl 1614913 polymorphisms are associated with genetic susceptibility of gastric cancer in Chinese Han population.
