目的 探讨在宫颈癌发生发展过程中蛋白质质谱的变化。方法 2005年6月至12月郑州大学第一附属医院采用表面增强激光解吸离子化飞行时间质谱仪,铜离子结合芯片检测宫颈上皮内瘤变(CIN)Ⅰ~Ⅱ级35例,Ⅲ级32例,宫颈癌Ⅰ~Ⅱa期39例,Ⅱb期以上26例以及71例健康女性血清,进行差异蛋白质的比较。结果 在宫颈癌发生发展过程中存在多种蛋白质的表达异常,其中具有分类意义的质荷比为8929.31、7930.52、9127.31、8141.01、7963.06、9280.63的6种蛋白质在宫颈浸润癌患者血清中低表达,含量由正常对照、CINⅠ~Ⅱ级、CINⅢ级、宫颈癌Ⅰ~Ⅱa、宫颈癌Ⅱb期以上,依次降低。结论 6种蛋白质与宫颈癌的发生发展密切相关,有可能成为与宫颈癌相关的一组生物学检测标志物。
Objective To characterize the changes of protein files in the initiation and development of cervical cancer. Methods Surface enhanced lazar desorption/ionisation-time of flight-mass spectrometry with immobilized metal affinity capture arrays-Cu was used to detect serum samples from 35 cases with cervical entraepithelial neoplasia Ⅰ - Ⅱ ,32 cases with CIN 111,39 cases with cervical cancer ( stage Ⅰ - Ⅱ a) , 26 cases with stage 11 b or more, and 71 control samples. All the samples were collected in the First Affiliated Hospital of Zhengzhou University from June 2005 to December 2005. The significant proteins were compared by t test. Results Many proteins/peptides had changed in cervical cancer initiation and development. Six proteins with mass to charge value of 8 929.31,7 930. 52,9 127.31,8 141. 01,7 963.06 and 9 280.63 appeared to be down regulated in patients with cervical cancer. The intensities orderly decreased from controls, CIN Ⅰ - Ⅱ , CIN Ⅲ to invasive cervical cancer in a level of 19 〈 0. 01. Conclusions These 6 proteins may be a group of biomarkers closely related to the origin and progress of cervical cancer.
