目的探讨MACC1(metastasis-associated in colon cancer-1)蛋白在肝细胞癌中的表达水平及其与患者预后之间的关系。方法免疫组化法检测MACC1蛋白在30例不同恶性程度的肝细胞癌中细胞分布特点;Western blot检测70例肝癌及其相对应的癌旁肝组织,30例正常肝组织中的MACC1蛋白的表达水平;随访分析不同患者的预后转归。结果免疫组化检测MACC1蛋白在肝细胞肝癌中阳性表达率为93.3%(28/30),Edmonson病理分级Ⅰ+Ⅱ和Ⅲ+Ⅳ级肝癌组织中MACC1核阳性表达率分别为26.3%、77.8%(5/19、7/9,P=0.031)。MACC1蛋白在肝癌组织、癌旁组织和正常肝组织中的相对灰度值分别为(1.33±1.17)、(0.59±0.57)、(0.003±0.001)(P=0.034)。高表达MACC1蛋白组和低表达组术后3年生存率分别为43.35%、73.45%(P=0.017)。结论 MACC1蛋白的表达异常与患者预后密切相关,联合检测MACC1蛋白的表达水平,对于指导临床治疗具有重要意义。
Objective To explore the expression of metastasis-associated in colon cancer-1(MACC1)protein in hepatocellular carcinoma(HCC)and its relationship with the prognosis of patients.Methods The expression of MACC1protein was immunohistochemically detected in 30cases of HCC with different degree of malignancy.The expression levels of MACC1protein in 70HCC tissues and their adjacent nontumorous liver tissues,and 30normal liver tissues were measured by Western blot.The patients were followed up and their prognosis was analyzed.Results Immunohistochemical staining showed that the positive expression rate of MACC1protein in HCC was 93.3%(28/30).The nuclear positive rate of MACC1protein was 26.3%and 77.8%in HCC with Edmonson gradesⅠ+ⅡandⅢ+Ⅳ,respectively(5/19,7/9;P=0.031).The relative gray value of MACC1protein was(1.33±1.17),(0.59±0.57),(0.003±0.001)in HCC tissues,adjacent nontumorous liver tissues and normal liver tissues,respectively(P=0.034).The 3-year survival rate was 43.35%and 73.45%in high MACC1expression group and low MACC1expression group,respectively(P=0.017).Conclusion The abnormal expression of MACC1 protein is closely related to the prognosis of HCC patients.Combined detection for the expression of MACC1protein,along with other relevant factors,is of great importance to guide clinical treatment.
