目的:对新疆贝母属8种药用贝母进行遗传多样性分析及评价。方法:采用ISSR分子标记技术综合分析8种药用贝母23份样品的遗传多样性。结果:16条引物共扩增出199条条带,其中多态性条带为195条,占97.99%。8种药用贝母遗传多样性水平为:Na=1.979 9,Ne=1.523 0,H=0.308 4,I=0.466 9,Gst=0.698 5,基因流(Nm*)为0.215 9,遗传一致度(I)和遗传距离(D)分别为0.747 4和0.301 4。表明8种药用贝母品种整体上有较高的遗传多样性,遗传变异主要存在于不同种间,基因交流程度比较有限。聚类结果显示,所有贝母均可明显区分开来,并聚为4组,遗传相似系数范围在0.58~0.99。结论:揭示了新疆贝母属8种药用贝母遗传多样性水平及亲缘关系,为其品种分类及种质保护提供理论依据。
Objective: To study the genetic diversity Methods: Inter-simple sequence repeat (ISSR) were diversity of the 23 individuals. Results: A total of 16 phic bands with the percentage of polymorphic bands served in all 8 species in Fritillaria L. (Na = 1. 979 of Fritillaria L. , 8 species of plants from Xinjiang. used to comprehensive analysis the level of genetic primers amplified 199 loci, including 195 polymor- at 97.99% Low levels of genetic diversity were ob- 9, Ne = 1. 523 0, H = 0. 3084, I = 0.4669, Gst = 0.698 5) gene flow(Nm. ) was 0.215 9, genetic identity (I) and genetic distance ( D ) were 0.7474 and 0.3014, respectively. The results indicate that the resources of 8 species in Fritillaria L. had high genetic diversity in general, Genetic diversity variation is mostly exists between species, the degree of gene flow among species is limited. Clustering analysis showed that the all Fritillaria L. could be distinc- tively classified into 4 groups with genetic similarity coefficient ranged from 0.58 - 0.99. Conclusion : The results indicate that the levels of genetic diversity and genetic relationship among 8 species in Fritil- laria L. , which could provide the varieties classification and species protection.
