目的 研究地昔帕明(desipramine,DMI)对大鼠海马神经元钾电流的影响,初步探讨其抗抑郁的离子通道机制。方法 酶解法急性分离单个大鼠海马神经元,应用全细胞膜片钳技术记录DMI对海马神经元钾电流的影响。结果 不同浓度DMI均可抑制海马神经元延迟整流钾电流(IK)和瞬时外向钾电流(IA)的影响,其中,100μmol·L^-1 DMI明显下移IK和IA电流-电压曲线,同时,DMI对IK的抑制率明显高于IA。结论 DMI对大鼠海马神经元外向钾电流具有明显的抑制作用,提示这可能是其神经保护的机制之一。
Aim To study the effect of desipramine (DMI) on the outward potassium currents in rat hippocampal neurons, and investigate its ion channels mechanisms of anti-depression. Methods Single hippocampal neuron of rat was obtained using acutely enzymatic isolation, and whole cell patch clamp technique was used to record the effect of DMI on outward potassium currents. Results DMI decreased the delay rectified potassium currents (IK) and the transient out- ward potassium currents (IA ) at different concentra-tlon, and signiticantly shitted the current-voltage curve of IK and IA downward at 100μmol· L^- 1 DMI. The in- hibition rate of DMI was higer on IK than on IA. Conclusions DMI has a significant inhibition on the outward potassium currents of rat hippocampal neurons suggesting that this might be one of the mechanisms of its neuroprotection.
