目的 探索组蛋白去乙酰化酶3(HDAC3)基因与小儿胶质瘤恶性分级以及预后的关系.方法 选取北京天坛医院2009年至2012年期间小儿神经外科病房收集的小儿胶质瘤标本(年龄≤15岁)70例,7例成人癫痫患者颞叶切除标本,Western blot以及RT-PCR方法检测HDAC3的蛋白表达和转录水平的表达,分析与胶质瘤病理分级的关系.相关分析检测HDAC3表达与患儿生存期的相关性.生存分析检测HDAC3表达与预后的关联性.结果 实验数据揭示在小儿胶质瘤患者中随着病理分级(WHO2007)的增加HDAC3表达也明显的增高(P<0.05),而在正常脑组织中几乎不表达.HDAC3表达与生存期呈负相关(P=0.046<0.05,Pearson Correlation=-0.387).增高的HDAC3表达与患儿差的预后相关联.结论 HDAC3基因表达水平与小儿胶质瘤的病理分级呈正相关,与患儿生存期负相关.
Objective To explore the relationship among histone deacety lases 3 (HDAC3),tumor grade and prognosis of children's gliomas.Methods The samples (n =70) were randomly collected from children with glioma and underwent microneurosurgery resection in the pediatric neurosurgery ward in Beijing Tiantan Hospital from 2009 to 2012.Normal brain tissues (n =7) as control groups were collected from adult patients with epilepsy who underwent temporal lobe resection in functional neurosurgery ward in Beijing Tiantan Hospital in 2012.Western blot and RT-PCR were used to detect the expression of HDAC3 and the relationship between the expression of HDAC3 and the tumor grade of glioma.Results Data showed that HDAC3 was significantly elevated in children's gliomas,following the glioma grade (WHO2007),with almost no expression in normal human brain tissues.Ectopic HDAC3 expression was correlated with a poor prognosis of children with glioma.Conclusion The level of HDAC3 expression was associated with tumor stages and survival time.
