目的探讨Ezrin蛋白在肺腺癌组织中的表达及意义。方法应用免疫组织化学染色检测92例肺腺癌及癌旁正常组织中的Ezrin蛋白表达,结合临床病理学指标及生存时间进行统计学分析。结果肺腺癌组织中的Ezrin阳性表达率明显低于癌旁正常组织(P〈0.01),Ezrin蛋白在组织分化程度较低、有转移以及高分期的肺腺癌组织中的阳性表达率明显降低(P值分别为0.023、0.003和0.001);癌组织中的Ezrin蛋白表达与患者的生存时间明显相关(Logrank=35.106,P〈0.01);Ezrin阴性表达是影响患者生存时间的独立风险因子(HR:0.408,95%CI:0.232-0.717,P=0.002)。结论Ezrin基因可以影响肿瘤的发生和发展,检测肺腺癌组织中Ezrin蛋白的表达对其预后评估有一定的价值,有望成为肺腺癌靶向治疗的分子靶点。
[Objective] To explore the role of Ezrin in tumor progression and prognostic evaluation oflung adenocarcinoma. [Methods] Immunohistochemistry for Ezrin protein was done to investigated the signifi-cance of Ezrin protein expression in 92 cases of lung adenocarcinoma. Additionally, the correlation betweenEzrin non-expression and the clinicopathological features of lung adenocarcinoma was evaluated by χ2 test andFisher's exact test, the overall survival rates were calculated by the Kaplan-Meier method, and the relation-ship between prognostic factors and patient over survival was also analyzed by the Cox analysis. [Results]Ezrin protein non-expression rate in lung adenocarcinoma was significant lower than the index in the adjacentlung tissues (P〈0.01). Ezrin non-expression rate was correlated with the differentiation, metastasis, and patho-logic stage (P =0.023, 0.003, and 0.001, respectively). Importantly, there was significant difference betweenEzrin non-expression rate and the overall survival rate (Log-rank= 35.106, P〈0.01). Further analysis by Coxproportional hazard regression model, Ezrin non-expression emerged as a significantly independent hazard factor for over survival of lung adenocarcinoma (HR: 0.408, 95%CI: 0.232 - 0.717, P=0.002). [Conclusion]Ezrin may affect the occurrence and development of lung adenocarcinoma. It might be a new attractivebiomarker for prognostic evaluation and molecular target therapy in patients with lung adenocarcinoma.
