提取与分离方法影响海兔口腔神经节蛋白质组的分离效果

选用丙酮沉淀法和直接裂解法分别提取离蓝斑背肛海兔(Notarcus leachii cinvsus Stimpson,NLCS)口腔神经节(Buccal Ganglion,BG)全蛋白质,发现采用直接裂解法所获得BG蛋白质种类多于丙酮沉淀法.在第一向等电聚焦电泳(IEF)中,分别选用pH为3.0—9.0,4.0~6.0和5.0~8.0的载体两性电解质,发现载体两性电解质pH范围为5.0—8.0时分离效果最好.使用Melanie 4 Trial软件统计BG蛋白质斑点数目约为290点,适于开展BG蛋白质组学研究.

英文摘要:

The total proteins ofbuccal ganglion from Aplysia were extracted by acetone precipitation or direct lysis. The results showed that more proteins could be obtained on the 2D-PAGE map by the latter approach. The carrier ampholytes of pH range 3.0 -9.0, 4.0 - 6.0, and 5.0 -8.0 were used in IEF respectively, it showed that the best separation effect could be obtained in pH range of 5. 0-8.0. And there are approximately 290 spots in the gel, these perfect datum indicated that this approach described here was suit for doing research about pro- teomics.
