目的:介绍一种改良的大鼠脑缺血再灌注模型。方法:40只SD雄性大鼠,按照Longa的线栓法制作脑缺血再灌注模型并进行改良,通过局部脑血流监测、神经功能缺损评分、CV染色和电镜等方法评价该模型的可靠性。结果:动物麻醉后一般只需15min左右即可完成栓线手术,术后大鼠大脑中动脉区域血流量下降,神经功能缺损明显,CV染色脑梗死区苍白,电镜显示缺血灶星形胶质细胞肿胀,神经元固缩坏死。结论:改良后方法优于传统方法,制备模型过程中,严密监测脑血流,严格控制脑血流量降至正常15%以下,夹闭翼腭突动脉,是提高造模成功率的关键。应用头端包被多聚赖氨酸并经熏香处理的尼龙线对血管损伤小。该改良的模型,缺血效果明确可靠,手术时间短,是理想的研究脑缺血的实验模型。
Objective:To introduce a modified rat model of middle cerebral artery occlusion(MCAO).Method:40 Sprague-Dawly male rats were used to induce focal cerebral ischemia reperfusion according to Longa's method.Focal cerebral blood flow(CBF),neurological deficit,cresyl violet(CV) stain and pathology were evaluated in all rats.Result:The operation proceeded about 15min after anaesthesia.Rats after operation presented severe neurological deficits companied with rCBF decreases.There were significant increases of brain water contents in ischemic brain regions.CV staining showed infarct areas and histological examinations revealed remarkable astrocyte swelling and neuronal necrosis.Conclusion:Modified methods is superior to traditional one.The cerebral blood flow reduction to 15% or less of baseline,ligation of pterygopalatine artery(PPA)are key factors in methods for rat model of cerebral ischemia reperfusion.Polysine head-coated nylon thread is less damage to blood vessel.The modified model is reliable,with the advantage of shortening operation time,is perfect for studying cerebral ischemia.
