目的:观察3’-大豆苷元磺酸钠对慢性心力衰竭大鼠心肌受磷蛋白(phospholamban,PLB)和Ca^2+-ATP酶(Ca^2+-ATPase,SERCA)mRNA表达的影响,探讨3’-大豆苷元磺酸钠抗心力衰竭的机制。方法:结扎大鼠冠状动脉前降支造成心肌梗死后慢性充血性心力衰竭(CHF)模型,采用实时荧光定量PCR技术观察。心肌PLB和SERCAmRNA表达以及3’-大豆苷元磺酸钠对其影响。结果:各给药组均可不同程度的下调PLBmRNA表达,上调降低SERCAmRNA表达,其中3’-大豆苷元磺酸钠0.4mg·kg。剂量组、地高辛组与模型组比较有显著性差异(P〈0.05);各给药组均能降低PLB/SERCA比值,地高辛组和3’-大豆苷元磺酸钠0.4mg·kg^-1、0.2mg·kg^-1剂量组与模型组比较差异具有显著性(P〈0.05,P〈0.01)。结论:3’-大豆苷元磺酸钠治疗能提高SERCAmRNA表达,降低PLBmRNA表达,改善PLB/SER-CA比值,这可能是3’-大豆苷元磺酸钠抗心力衰竭的机制之一。
Objective :To observe the effect of 3'-daidzein sulfonate sodium on the expression of PLB and SERCA mRNA in chronic heart failure(CHF) rats and the mechanism of 3'-daidzein sulfonate sodium against heart failure. Methods:The model of CHF was induced by ligating the anterior descending branch of the left coronary artery. The rats were at random divided into six groups:sham operation group,model group,digaoxin group,three treatment groups treated with 0. 4mg · kg^-1 ,0. 2mg · kg^-1 and 0. 1 mg · kg^-1 dosage of 3'- daidzein sulfonate sodium. After administration for 4 weeks. The effect of 3 '- daidzein sulfonate sodium in treatment on the expression of PLB and SERCA mRNA was observed by RT - PCR. Results : All the treated groups could decrease the expression of PLB mRNA and increase the expression of SERCA mRNA in different degree, and there was significant difference between the 3'-daidzein sulfonate sodium 0.4mg · kg^-1 dosage treated group, digaoxin group and model group (P 〈 0.05 ) ;All the treated groups could decrease ratio of PLB/SERCA. Compared with model group, digaoxin group and the 3'- daidzein sulfonate sodium 0. 4mg · kg^-1,0. 2mg · kg^-1 dosage treated group had significant difference (P 〈 0. 05, P 〈 0.01 ). Condusion:Glucoperiploeymar in treatment could increase the expression of SERCA mRNA, decrease the expression of PLB mRNA and improve the ratio of PLB/SERCA, and it might be one of the mechanism of 3'- daidzein suffonate sodium in against heart failure.
