目的:研究宿主因素对毛囊重建的影响即探讨不同种系小鼠(免疫功能缺陷及正常小鼠)作为宿主时毛发再生情况。方法:将GFP新生鼠的表皮、真皮单细胞以一定比例注射到C57和裸鼠体内。诱导形成异位毛发,并于移植后2、4、7、14、18、21 d取材进行观察,随后每隔3 d取材,直至观察到所形成毛发重新进入生长期。结果:两种小鼠体内均可形成新的毛囊,新生毛囊在诱导阶段大致相同。但C57鼠新生毛囊在18 d进入退行期,而裸鼠在21 d。C57鼠新生毛囊在33 d重新进入生长期,裸鼠则在39 d。HE切片见再生毛囊结构完整,冷冻切片荧光检测见明亮绿色荧光。细胞以不同的比例移植后发现,C57鼠体内毛囊重建效率高于裸鼠,且二者的真表皮细胞的最佳比例不同,单一细胞移植至两种鼠体内时均无毛发形成。结论:新生鼠真表皮细胞注射到不同种系小鼠体内均能形成新的毛囊,不局限于免疫功能缺陷宿主,但宿主因素对毛囊重建也会造成一定影响,所形成毛囊的周期、毛囊重建效率及形成的毛发性状不同。
Objective To investigate hair regeneration among different species of mice as the host. Methods Dermal and epidermaI cells isolated from newborn GFP mice skin.Patch assay was made with dermal and epidermal cells isolated from newborn GFP mice skin at a specific ratio on days 2, 4, 7, 14, 18, and 21 days. The skin was cut off from the injected site, and then hair formation was observed, then skin of the injected site was cut off every 3 d until the hair follicle re-enter anagen. Results New hair formation was seen in two species of host, and the phase of hair follicles induction was roughly the same. But the newly formed hair follicles in C57 grew into catagen on day 18, and those on nude mice did on day 21. The newly formed follicles in C57 re-ran anagen phase on day 33, and those on the nude mice did on day 39. The completely developed structure of hair follicles was observed with paraffin section and HE staining. The newly formed follicles showed bright green fluorescence. The ratio and number of cell components were varied, and the efficiency of hair foloicle regeneration in C57 mice was higher than in the nude ones. Neither epidermal cells nor dermal cells transplanted alone formed hair follicle in the two species of mice. Conclusion Newborn mice skin cells can form new hair follicles within different species after implantation, which is not specific to immunoincompetent mice such as the nude (nu/nu) mutant. However, the hosts may also render influence on the reconstruction of hair follicles and the regenerated hair follicles have different cycles and features.
