目的评价肝肺综合征(HPS)大鼠肺组织膜联蛋白A2(ANXA2)表达的变化。方法健康sD大鼠40只,雌雄不拘,3~4月龄,体重220~250g。采用随机数字表法,将其分为3组:对照组(c组,n=10)、假手术组(s组,n=10)和肝肺综合征组(HPS组,n=20)。HPS组采用胆总管结扎法建立肝肺综合征模型;c组不做任何处理;S组仅开腹暴露胆总管,但不结扎。术后5周时,处死大鼠,取肺组织,采用RT—PCR法检测ANXA2和平滑肌肌动蛋白α(SM-α-actin)的mRNA表达,采用Westernblot法检测ANXA2和SM—α—actin的蛋白表达。结果与C组比较,S组肺组织ANXA2和SM—α—actin的mRNA及其蛋白表达差异无统计学意义(P〉0.05);与s组比较,HPS组肺组织ANXA2和SM—α-ctin的mRNA及其蛋白表达上调(P〈0.05)。结论大鼠肝肺综合征时肺组织ANXA2表达上调。
Objective To evaluate the changes in the expression of annexin A2 (ANXA2) in lung tissues in rats with hepato-pulmonary syndrome. Methods Healthy 3-4-month-old Sprague-Dawley rats of both sexes, weighing 220-250 g, were randomly divided into 3 groups: control group (group C, n = 10), sham operation group (group S, n = 10) and hepatopulmonary syndrome group (group HPS, n = 20) . The rats were anesthetized with intraperitoneal 3 % pentobarbital sodium 0.1 ml/100 g. Hepatopulmonary syndrome was produced by chronic ligation of the common bile duct. After 5 weeks, the rats were sacrificed and lungs were removed for determination of ANXA2 and smooth muscle actin a (SM-α-actin) mRNA and protein expression in lung tissues by RT-PCR and Western blot, respectively. Results There was no significant difference in the expression of ANXA2 and SM-α-actin mRNA and protein between groups C and S ( P 〉 0.05). The expression of ANXA2 and SM-α-actin mRNA and protein was significantly higher in group HPS than in groups C and S ( P 〈 0.05). Conclusion The expression of ANXA2 in lung tissues is up-regulated in rats with hepato-pulmonary syndrome.
