目的:探讨骨髓基质干细胞(BMSCs)移植与中药合用治疗糖尿病足溃疡的疗效及血管内皮生长l因子(VEGF)的表达情况。方法:取新西兰兔32只,建立2型糖尿病足溃疡模型,造模后按随机数字表法分为4组:对照组(A组),中药治疗组(B组),BMSCs移植治疗组(c组),BMSCs移植联合中药治疗组(D组),每组8只。治疗15d,观察各组溃疡创面组织切片染色及进行VEGFmRNA和蛋白检测。结果:与A组比较,B、C组溃疡愈合较好,创面组织中VEGFmRNA和蛋白表达增高(P〈0.05),D组溃疡愈合迅速,创面组织中VEGFmRNA和蛋白显著增高(P〈O.01)。结论:BMSCs移植配合中药治疗糖尿病足溃疡可促进溃疡创面的愈合,可能机制为其上调了局部VEGF基因和蛋白的表达。
Objective: Explore the efficacy of bone marrow stromal cell transplantation combined with traditional Chinese medicine treatment of diabetic foot ulcer and the expression of vascular endothelial growth factor (VEGF). Methods: 32 New Zealand rabbits were made type 2 diabetic foot ulcer model, and according to the random number table which were divided into four groups: control group (A group), traditional Chinese medicine treatment group (B group), bone marrow stromal cells transplantation group (C group), bone marrow stromal cells transplantation combined with traditional Chinese medicine treatment group (D group), 8 rabbits in each group. After 15 days of treatment, we observed each group ulcer wound tissue sections, and VEGF mRNA was detected. Results: Compared with A group, B group and C group are better to heal and VEGF mRNA and protein expression were increased in wound tissue (P〈0.05). D group ulcer healing rapidly and VEGF mRNA and protein expression were significantly increased in wound tissue (P〈0.01). Conclusion: BMSCs transplantation with Chinese medicine treatment of diabetic foot ulcer can promote wound healing of ulcer, and the possible mechanisms for its maybe raise local VEGF gene and protein expression.
