目的通过以“超广谱”、高水平耐药的摩氏摩根菌(Morganellamorganii)KL-225为模式生物建立筛选模型。方法分别在培养基中加入青霉素、头孢吡肟、链霉素、环丙沙星、左氧氟沙星、交沙霉素、杆菌肽、黏菌素、利福平、磺胺嘧啶等,以此作为耐药菌的耐药筛选指标,从养殖池塘水体和病死的鲍鱼体的菌群中筛选能够耐受上述药物的新型摩氏摩根菌。结果筛选到广谱耐药摩氏摩根菌,构建细胞水平的新型抗菌药物筛选模型,对本所重点实验室化合物库进行初筛及复筛得到阳性化合物H1,其对广谱耐药摩氏摩根菌KL-225和摩氏摩根标准菌的MIC值都是1.59gg/mL,检测了H1的抗菌谱尤其是阴性耐药抗菌谱,考察了H1的细胞毒作用。结论通过以“广谱耐药”的摩氏摩根菌为模式菌株建立的筛选模型可用于抗耐药菌药物的发现。
Objective A screening model has been builded by a high level multidrug resistant bacterium of Moganella morganii KL-225 as a model organism. Methods Penicillin, cefepime, streptomycin, ciprofloxacin, levofloxacin, josamycin, bacitracin, polymycin, rifampin, sulfadiazine such as drug-resistant screening indicators was added in the medium to-find resistant bacteria. Thus, the new Morganella morganii strain which can tolerate all above drugs was isolated from dead abalones. Results New antimicrobial drug screening model is builded at the cellular level through XDR Morganella morganii as a model organism. The positive compound H1 was gotten through screening and rescreening laboratory compound library, its MIC values of multi-drug resistant bacteria Morganella morganii KL-225 and Morganella morganii standard bacteria are 1.59ktg/mL both, its antibacterial spectrum especially gram-negative resistance was detected, its cytotoxic effect was investigated. Conclusion The screening model whose mode organism is "XDR"Morganella morganii can be used to discover the anti-drug-resistant drug.
