研究莱菔硫烷(sulforaphane,SFN)对内质网相关蛋白Bip/GRP78和caspase-12蛋白表达的影响,探讨其诱导人肝癌HepG-2细胞的凋亡机制.采用Western Blot方法检测人肝癌HepG-2细胞内Bip/GRP78蛋白的表达,以及采用流式细胞仪检测caspase-12蛋白的表达.10、20、40μmol/L的SFN作用于人肝癌HepG-2细胞48 h后可显著上调HepG-2细胞内Bip/GRP78和caspase-12蛋白的表达.SNF可能通过上调HepG-2细胞内Bip/GRP78和caspase-12蛋白的表达启动内质网途径诱导肝癌HepG-2细胞的凋亡.
To study the effect of sulforaphane(SFN) on the endoplasmic reticulum-associated protein expression of Bip/GRP78 and caspase-12 in human HepG-2 cells and to explore the inducing apoptotic mechanism of SFN on HepG-2 cells.Western Blot assay was adopted to detect the expression of Bip/GRP78 protein and Flow cytometry was used to detect the expression of caspase-12 protein.After treated by SFN for 48h,different dosages of SFN could remarkably up-regulate the expression of Bip/GRP78 and caspase-12 in HepG-2 cells.SFN can up-regulate the expression of Bip/GRP78 and caspase-12,by which induce apoptosis of HepG-2 cells through the endoplasmic reticulum pathway.
