利用高效毛细管法(HPCE)对人工倾斜的中林46杨(Populus×euramericana(Dode)Guiner CL.Zhonglin-46)树干形成层内源激素进行测定。结合自身试验材料的特殊性,优化样品前处理过程,选择符合自身材料的最佳电泳条件,探索出测定树干形成层5种内源激素赤霉素(GA3)、吲哚-3-乙酸(IAA)、吲哚-3-丁酸(IBA)、玉米素(Z)和脱落酸(ABA)的方法。最佳条件为:电压16 kV、柱温20℃、缓冲液pH 9.25、SDS浓度50mmo.lL-1。5种内源激素的相关系数R2为0.978 6~0.999 9,回收率为93%~105%,变异系数≤3.93%。
The endogenous hormones in inclined stem cambium of poplar(Populus×euramericana(Dode) Guiner CL.Zhonglin-46) were determined by high-performance capillary electricity(HPCE).The five endogeneous hormones such as gibberellic acid(GA3),indole-3-Acetic acid(IAA),indole-3-butyric acid(IBA),zeatin(Z) and abscisic acid(ABA) in cambium of poplar stem were separated by optimizing the pre-processing procedure and the electrophoretic conditions suitable for the particularity of the material.The best conditions are as follows:voltage 16 kV,column temperature 20℃,the pH 9.25 for electrolyte solution,and concentration of SDS is 50 mmol·L-1.The corresponding coefficient of five kinds of endogenous hormones reach from 0.9786 to 0.9999.The recoveries of hormones vary from 93% to 105%,and the coefficients of variation are less than 3.93%.
