在构建乙烯利刺激条件下巴西橡胶树(Hevea brasiliensis)胶乳差异表达cDNA消减文库的基础上,克隆了胶乳HbUEP(Hevea brasiliensis ubiquitin extension protein)基因cDNA全长序列(GenBank accession No.EF120638)。结构分析表明,该基因cDNA全长771bp,拥有一个468bp的开放阅读框架、77个核苷酸的5'UTR和226个核苷酸的3'UTR,编码156个氨基酸;Southern blot检测结果显示,HbUEP基因在巴西橡胶树基因组中属低拷贝基因。HbUEP基因的表达受到乙烯利的调节。在施用乙烯利后的24h内,12h收集的胶乳中HbUEP基因表达最强,对照和6h收集的胶乳中表达最弱。乙烯利刺激可以促进巴西橡胶树胶乳增产。这一结果表明,HbUEP基因可能参与了乙烯利刺激巴西橡胶树胶乳增产的分子调控。
Based on employing differentially ethphon-induced expressed cDNA subtracted library resulted from latex in rubber trees(Hevea brasiliensis), HbUEP gene, an ubiquitin extension protein gene from latex in H.brasiliensis (GenBank accession No.EF120638) was cloned and sequenced. HbUEP gene cDNA had 771 bp nucleotides, comprising 226 bp-length 3'UTR, 77 bp-length 5'UTR and a 468 bp-length open reading frame encoding 156 amino acid peptide. Southern blotting analysis showed that this gene was a kind of low-copy genes in H. brasiliensis genomic. Within 24 h after application of ethphon HbUEP gene lowly expressed in both control and 6 h-sampled latex,and strongly in 12 h-sampled latex, showing that this gene expression could be regulated by ethphon and ethphon could increase the latex yield in H.brasiliensis. It is suggested that HbUEP gene may be involved in the regulation of ethphon-induced high latex yield in H.brasiliensis.
