目的对白及多糖衍生物胆甾醇取代度的测定方法进行研究。方法采用HPLC法、UV法和核磁共振法(1H-NMR)对合成的胆甾醇琥珀酰基白及多糖进行取代度的测定。结果 HPLC法、UV法、核磁共振法测定胆甾醇取代度的结果分别为3.84%、3.82%、4.26%。结论 3种测定方法都有可行性,HPLC法、UV法比核磁共振法测定胆甾醇值略低,但偏差不大,各有优缺点,可根据对试验结果准确度的要求选择较为合适的测定方法。
Objective To compare three methods for determination of the degree of substitution of polysaccha- ride derivatives. Methods High performance liquid chromatography (HPLC), UV visible spectrophotometric method and nuclear magnetic resonance spectroscopy (NMR) were used to determine the degree of substitution in synthethic cholesteryl succinyl of sugar (CHSB) . Results The degree of substitution were 3. 84%, 3. 82% and 4.26% respectively. Conclusion All the three methods are applicable. HPLC and UV visible speetrophotometry yielded slightly lower values than nuclear magnetic resonance method, but deviation is small. Each has its advantages and disadvantages. The selection of a method depends on the requirements of accuracy of experimental results.
