应用反相高效液相色谱和质谱联合使用手段,从沼水蛙皮肤分泌液中分离到一种新型的Temporin家族成员—Temporin-HZ,经过Edman降解氨基酸序列分析和串联质谱研究,Temporin-HZ的一级结构最终确定为Phe-Leu—Pro-Leu-Leu-Phe-Gla-Ala-Ile-Ser-His-Leu-Leu-NH2。抗菌活性分析表明,Temporin-HZ对大肠杆菌和白色念珠菌生长无明显抑制,但对金黄色葡萄球菌有抗菌作用。
A new temporin-HZ was isolated from the skin secretion of Hylarana guentheri by reverse phase HPLC and mass spectrometry analysis. The primary structure of temporin-HZ (Phe-Leu-Pro-Lcu-Lcu-Phe-Gla-Ala-Ile-Ser-His-Leu-Leu-NH2) was identified after Edman degradation amino acid sequencing and ms/ms Q-TOF analyzing. The tempofin-HZ had moderate activity against Staphylococcus aureus and no activity against Escherichia coli and Candida albicans.
