脑胶质瘤由于浸润性生长的特点以及存在血脑屏障(BBB),用于胶质瘤术前、术中定位诊断的成像效果不佳,常规的手术后放、化疗效果不佳,因此寻找可以穿越血脑屏障并靶向到肿瘤上的显像剂以及新的胶质瘤治疗方法尤为重要。近年来发现一种多肽Angiopep-2,能够通过高表达于血脑屏障和胶质瘤的低密度脂蛋白受体相关蛋白(LRP)介导,靶向血脑屏障和脑胶质瘤细胞,因此可以用Angiopep-2来修饰纳米递药和成像系统,以提高药物疗效和成像效果。本文综述了Angiopep-2的材料特点、血脑屏障的生理特征以及Angiopep-2修饰的纳米递药系统和纳米成像系统在胶质瘤诊疗中的应用进展。
The poor quality of images of glioma which are used preoperatively and intraoperatively is due to the invasive growth of brain gliomas and the blood-brain barrier(BBB), so brain gliomas has not got good therapeutic effect by subsequent radiotherapy and chemotherapy after surgery. Therefore, new imaging agents and therapeutic methods targeting both BBB and glioma are very imperative. Recent years, a new polypeptide named Angiopep-2, a ligand connected with low density lipoprotein receptor related proteins(LRP) which are over expressed on both BBB and glioma, is used to improve the quality of images during surgery and therapeutic effects of nanoparticle delivery system. In this review, the characteristics of Angiopep-2, physiological features of BBB and the applying of nanoparticle delivery system decorated with Angiopep-2 are presented.
