目的探讨电针对SD大鼠骨性关节炎模型血液及关节液中COX2、iNOS、NO表达的影响。方法成年SD大鼠180只,抽签法随机分为正常组、实验组,实验组采用4%木瓜蛋白酶双膝关节腔注射复制骨性关节炎模型,抽签法随机分为模型组、对照组、实验1组、实验2组、实验3组,对照组采用关节腔注射透明质酸钠,实验组采用电针干预,检测关节液中COX2、iNOS、NO含量,光学显微镜观察关节软骨的形态结构。结果血液及关节液中COX2、iNOS、NO,各实验组均高于正常组(P〈0.05,P〈0.01),对照组、实验2组、实验3组明显低于模型组(P〈0.05,P〈0.01);关节软骨改变的分数,各实验组均高于正常组(P〈0.01),对照组、实验2组、实验3组明显低于模型组(P〈0.01)。结论电针能有效抑制关节液中COX2、iNOS、NO表达,延缓关节软骨的退变。
Objective To explore the effect of electroacupuncture on the expression of COX2,iNOS and NO in blood and synovia of SD rat models of osteoarthritis.Methods 180 SD rats were randomly divided into 2 groups.Rabbits in the experimental group were replicated into knee osteoarthritis model by intra-articular injection of 4% Caroid,and they were randomly divided into model group,control group,experimental group 1,experimental group 2 and experimental group 3.Rabbits in the control group were treated with intra-articular injection of Sodium Hyaluronate,and those in the experimental group were treated with electroacupuncture.The levels of COX2,iNOS and NO in synovial were detected and morphology of articular cartilage was observed by light microscope.Results In each experimental group,the levels of COX2,iNOS and NO in blood and synovial were higher than those of normal group(P〈0.05,P〈0.01),and the levels of COX2,iNOS and NO in control group,experimental group 2 and experimental group 3 were lower than those of model group respectively(P〈0.05,P〈0.01);the amplitude of variation of articular cartilage score in each experimental group was bigger than that of normal group(P〈0.01),and the amplitude of variation in control group,experimental group 2 and experimental group 3 were smaller than that of model group respectively(P〈0.01).Conclusion Electroacupuncture can inhibit the expression of COX2,iNOS and NO in synovial,thereby to delay the degeneration of articular cartilage changes.
