目的:研究氯化铝对大鼠基底前脑内侧隔核APP蛋白表达的影响.方法:选用雄性Wistar大鼠20只,随机分为老龄组(18个月)、老龄给药组(自18个月始给予腹腔内注射氯化铝至21个月),21个月后进行Morris水迷宫训练及测试,观察大鼠的行为学改变.然后采用免疫组织化学SP法检测大鼠内侧隔核APP蛋白表达的变化情况.结果:老龄给药组大鼠逃避潜伏期与老龄组相比时间增加(P<0.05);老龄给药组内侧隔核APP蛋白阳性反应物的平均光密度值与老龄组相比显著增高(P<0.05).结论:老龄给药组大鼠内侧隔核APP蛋白表达明显高于老龄组,说明长期接触氯化铝可能导致基底前脑内侧隔核神经元APP蛋白表达或聚集增加.
Objective: To explore the effect of aluminum chloride on APP in medial septum of aged rats. Methods: Twenty male wister rats were randomly divided into aged group ( 18 months) and aged group with aluminum chloride ( intraperitoneal injection aluminum chloride from 18 months to 21 months). The quantitative changes of APP in medial septum of wistar rats were studied with immunohistochemical SP method. Results: The escape latency period of aged group with aluminum chloride was increased compared with that of aged group ( P 〈 0. 05 ). The optical density value of aged group with aluminum chloride was increased compared with that of aged group ( P 〈 0. 05 ). Conclusion : The level of APP expression is increased in medial septum of aged group with alumi- num chloride, so long-term exposure to aluminum chloride can result in increase for APP expression and aggregation in neurons of basal forebrain.
