为研究转脂蛋白(lipid transfer protein,LTP)作为抗菌肽在油菜防御反应中的作用,利用生物信息学的方法,从油菜中筛选获得了一个转脂蛋白类抗菌肽基因,命名为Bn LTP1。Bn LTP1基因长度为93bp,编码31个氨基酸残基。抗菌肽Bn LTP1分子量约为3.4k Da,其序列中的4个半胱氨酸形成两个分子内二硫键,对于稳定抗菌肽的结构起到至关重要的作用。通过overlap PCR的方法对该基因进行体外人工合成,并克隆到p ET30a/His-EDDIEGFP表达载体上进行体外原核表达。抑菌活性实验检测发现,Bn LTP1对革兰氏阳性菌、革兰氏阴性菌和酵母菌都有较强的抑菌活性;对植物病原真菌(核盘菌、灰霉菌和稻瘟病菌)等也具有抑菌活性。
Lipid transfer protein( LTP) play an important role in plant defense. In this study,Bn LTP1 gene was cloned from Brassica napus L. It harbored 93 bp,encoding 31- amino acids with molecular weight of 3. 4 k Da.The gene was synthesized artificially by overlap polymerase chain reaction( PCR) and cloned into the prokaryotic expression vector ET30 a / His- EDDIE- GFP. Its peptide Bn LTP1 was expressed in Escherichia coli BL21. Antimicrobial activity demonstrated that Bn LTP1 exhibited strong antimicrobial activity against Gram- positive bacteria,Gram- negative bacteria and yeast. Bn LTP1 also had strong antifungal activity against several plant pathogenic fungi,including Sclerotinia sclerotiorum,Magnaporthe oryzae and Botrytis cinerea.
