在单因素实验的基础上,用正交实验法确定了超声波辅助提取一点红花色素苷的最佳工艺条件.结果为:按液料比80:1用80%酸性乙醇于70℃水浴浸泡30min后,采用240W超声波功率浸提25min.同时比较了超声波提取法与常规提取法的提取效果,在各自的最佳工艺条件下,超声辅助法和常规溶剂浸提法的优化工艺都能较充分地提取一点红中的花色素苷生物活性成分,但超声辅助提取的时间明显短于常规溶剂浸提法.一点红花色素苷稳定性的实验表明,一点红花色素苷对酸性条件敏感;提取液在低于80℃时是稳定的,对高温敏感;三氯化铁、蔗糖和柠檬酸、苯甲酸钠和山梨酸钾对一点红花色素苷有降解作用.
The optimal processing conditions for ultrasonic extraction of anthocynin from Emilia sonchifoli were obtained by single factor and orthogonal tests. The results show: 240 W ultrasonic powder, 80% (volume fraction) ethanol as solvent,the ratio of liquid to material is 80:1 ,extracting once for 25 mins at 70 ℃. Ultrasonic extraction has superiority over traditional methods. Ultrasonic extraction has advantages of shorter duration and less energy consumption. Moreover, the anthocynin is stable at temperature below 80 ℃ , but sensitive to acid pH and boiling water. Sucrose, citric acid, sodium benzoate and potassium sorbate can degrade the anthocynin.
