硬组织切片技术在血管支架研究中的应用

目的 采用改进的硬组织切片技术观察血管成形术后的支架内再狭窄情况。方法 表面改性的血管支架对中国小型猪进行经皮冠状动脉成形术(PTCA),植入3个月后取出固定,对硬组织切片的组织包埋、切片及染色的条件进行探索和改进。结果 通过改良方法制备的血管支架切片,能保持原有的组织形态结构,可进行HE及免疫组织化学染色。染色后组织结构完整,细胞形态清晰,可进行后续的病理学研究。结论 改良后的硬组织切片技术能成功观察和评价血管成形术后的支架内再狭窄情况,为支架内再狭窄的机理研究提供了较为实际的方法,适合推广。

英文摘要:

Objective To observe the progress of in-stent restenosis after percutaneous transluminal coronary angioplasty (PTCA) with the improved method of hard tissue slicing. Methods The Chinese pigs after PTCA were implanted with the superficially modified intravascular stents. After 3 months the intravascular stents were taken out. The conditions of embedding, cutting and staining of hard tissue slicing were explored and improved. Results The improved techniques in hard tissue slicing of intravascular stents could keep the morphosis of tissues and easily stain with HE and immunohistochemistry. The stained hard tissue slicing had an intact tissue structure and clear cell form, which could be used for the successive pathological research. Conclusion The factors which need to be improved in the analysis of pathology are been determined. The improved techniques in hard tissue slicing can successfully observe and evaluate the progress of in-stent restenosis after PTCA, which provides a more actual research method of mechanism of in-stent restenosis and is suitable to be spread.
