观察3T3-L1脂肪细胞诱导分化过程中UCP4基因mRNA表达水平的变化,探讨TNF-α对成熟脂肪细胞中UCP4基因表达水平的调节作用。研究发现UCP4基因在3T3-L1细胞分化过程中表达逐渐下调,其机制可能有利于脂肪细胞的分化成熟和脂质积聚,与肥胖发生有关;TNF-α能够上调成熟脂肪细胞中UCP4基因的表达,其效应总体趋势上呈现时间反应性特征。
The change of UCP4 gene expression during differentiation of 3T3-L1 preadipocyte was observed and the regulative role of tumor necrosis factor-α (TNF-α) in UCP4 gene expression in mature adipocytes was explored. The results showed that down-regulation of UCP4 gene mRNA expression during differentiation of 3T3-L1 preadipocyte may contribute to the differentiation, maturing and adipogenesis of adipocytes and that UCP4 gene may be related to the etiology of obesity. TNF-α can upregulate the mRNA expression of UCP4 gene in the mature adipocytes. The effect of TNF-α on UCP4 gene expression is generally of time-responsive correlation.
