本研究利用RNA干扰技术研究亚洲璃眼蜱p36基因在其生长、发育、繁殖方面的生物学功能。通过注射p36基因的dsRNA成功实现了该基因的表达沉默,干扰组表达量下降了99%。蜱p36基因沉默后,24 h的吸附率下降了50%,饱血率下降了55.5%,卵孵化率下降了100%。结果表明p36基因在亚洲璃眼蜱吸血和繁殖中具有重要作用。
In this study,the functions of p36 gene of Hyaloma asiaticum(Ha-p36) in growth,development and reproduction were studied using RNA interference(RNAi).By the injection of double-strand RNA(dsRNA) of Ha-p36,gene silencing was confirmed by real-time PCR,which showed the expression amount of Ha-p36 gene decreased 99% after RNAi.Comparing with the control,the tick attachment rate,engorgement rate and egg hatching rate decreased 50%,55.5% and 100% in the RNAi group,respectively.The results indicated that Ha-p36 might play an important role in the growth and reproduction of H.asiaticum.
