目的:探讨丹参素钠对大鼠肝癌细胞及自杀基因旁观者效应影响。方法:丹参素钠与丙氧鸟苷(GCV)分别或共同作用于大鼠肝癌CBRH7919的tk-细胞,以及含5%tk+细胞的tk+和tk-混合细胞,MTT检测各组存活率,两两比较各组存活率的差异,用q值分析药物与自杀基因系统联合的相互作用是否为协同性。结果:丹参素钠12.5、25、50、100 mg/L作用于CBRH 7919细胞72h存活率分别为87.8±12.4%、46.0±6.4%、20.2±4.6%、8.9±1.2%;IC50为24mg/L。10mg/L和50mg/L组的丹参素钠联合5%tk+/GCV组的细胞存活率比单纯丹参素钠或单纯自杀基因组明显低,两两比较差异有统计学意义(P〈0.05),q值均〉1.15。结论:丹参素钠有明显抑制癌细胞生长作用,并能协同性增强tk/GCV系统对癌细胞的杀伤作用,增强自杀基因旁观者效应。
Objective: To explore the effects of Danshensu Sodium on rat hepatocarcinoma cells and the bystander effect of herpes simplex virus thymidine kinase gene/ganciclovir (HSV1 -tk/GCV). Methods: Rat hepatocarcinoma cell line CBRH7919/tk^- , CBRH7919/tk^+ and the 5% tk^+ mixed cells were treated with diverse concentrations of Danshensu Sodium and GCV separately, or Danshensu Sodium plus GCV ( n = 3 - 4). The survival rate of the cells in each group was examined by MTT method, and q -value analysis was used to estimate the synergistic effect of Danshensu Sodium on the suicide gene system. Results: After the treatment of 12.5, 25, 50, 100 mg/L Danshensu Soudium for 72 h, the survival rate of CBRH 7919/tk^- was 87.8 ± 12.4%, 46.0 ± 6.4%, 20.2 ± 4.6% , 8.9 ± 1.2% , respectively. IC50 was 24 mg/L. Both of the survival rate in groups of 10mg/L and 50mg/L Danshensu Sodium + 5% tk^+/GCV are significantly lower than that in groups of relatively concentration, of Danshensu Sodium and 5% tk^+/GCV group( P 〈 0.05 ) , q values 〉 1.15. Conclusion: Danshensu Sodium can inhibit the proliferation of hepatocarcinoma cells significantly, and can synergistically enhance the killing and bystander effect of tk/ GCV system.
