目的:观测1~5型重组腺相关病毒(adeno—associated virus,AAV)对牙龈、牙槽骨和牙周膜细胞的转染效率。方法:在小鼠龈下注射采用绿色荧光蛋白(green fluorescent protein,GFP)标记的1~5型AAV,术后21d收集标本,制作冰冻切片,荧光显微镜观测GFP标记的阳性细胞,计算各型病毒对牙周组织的转染效率。结果:AAV5和AAV2对牙周组织的转染效率较高,AAV3、AAV1和AAV4较低;AAV2对牙龈组织的趋向性高,而AAV3对牙周膜的趋向性高。结论:不同血清型的AAV对不同牙周组织的转染效率有显著差异。
Objective:To observe in vivo transfection efficiencies of 1-5 serotype recombinant adeno-associated viruses (AAVs)on gingival, alveolar, and periodontal ligament cells. Methods:1-5 AAVs marked with green fluorescent protein(GFP) were injected under the mouse gingivae, and samples were collected 21 days after injection. Transfection efficiencies were observed under fluorescent microscopy according to the measurements of GFP positive cells. Results:AAV5 and AAV2 had higher transfection efficiencies than AAV3, AAV1 and AAV4. The trendencies of AAV2 to gingival cells and AAV3 to periodontal ligament ceils were much higher than orthers. Conclusions:Different AAVs have significantly different transfection efficiencies on different periodontal tissues.
