建立了同时测定百尾参中obtucarbamate A和obtucarbamate B两种酰胺类化合物含量的高效液相色谱分析方法,色谱柱为Agilent ZORBAX SB-C18(250 mm×4.6 mm,5μm),以甲醇-0.2%乙酸水(30∶70)为流动相,检测波长256nm,流速1mL·min-1,柱温30℃.结果表明:obtucarbamate A和obtucarbamate B分别在3~96μg·mL-1和1~32μg·mL-1范围内浓度与峰面积呈良好线性关系,其平均回收率分别为99.23%和98.94%,RSD分别为1.11%和1.57%.该方法操作简单,专属性强,灵敏度高,适用于百尾参中酰胺类化合物的含量测定.
An ananlytic method using high-performance liquid chromatograph(HPLC)for determination obtucarbamate A and obtucarbamate B of Disporum cantoniense was established.The determinination was performed on Agilent ZORBAX SB-C18(250mm×4.6mm,5μm)column,methanol-0.2% acetic acid aqueous solution(30∶70)as mobile phases at a flow rate 1.0 mL·min-1.The detection wavelength was256 nm and the column temperature was 30 ℃.The results showed that the calibration curves of obtucarbamate A and obtucarbamate B had good linearity in the range of 3~96μg·mL-1 and 1~32μg·mL-1,respectively.The average recoveries were 99.23 %(RSD=1.11 %)and 98.94 %(RSD=1.57 %),respectively.The method is simple,good specialization,high sensitive,and could be used to simultaneously determine amides of Disporum cantoniense.
