研究了曙红Y(EOSY)与牛血清白蛋白(BSA)作用的荧光光谱和紫外可见吸收光谱特征,实验证明EOSY具有较强地猝灭BSA荧光强度的能力,分别用Stern-Volmer方程、Lineweaver-Burk方程和热力学方程等处理试验数据,得到了25℃时反应的结合常数为3.601×10^5L·mol^-1,反应的△H^θ,△G^θ和△S^θ分别为-20.66,-31.70kJ·mol^-1和37.06J·K^-1,为研究在人体生理条件下EOSY与BSA的结合方式、EOSY对BSA构象影响和细胞染色机理等提供了重要信息。
The binding of eosin Y to bovine serum albumin (BSA) was studied by fluorescence spectroscopy and ultraviolet spectrum. It was shown that this compound has a quite strong ability to quench the fluorescence from BSA. After analyzing the fluorescence quenching data according to Sterm-Volmer equation, Lineweaver-Burk equation and thermodynamic equation at 298. 15 K, the binding constant (KLB = 3. 601 ×10^5L · mol^-1 ) and thermodynamic parameters (△H^θ, : - 22.66 kJ ~ mol 1, △G^θ: -31.30 kJ · mol 1 and △S^θ: 36. 32 J · K^-1)were obtained, which provide important information for researching the configuration modification of BSA caused by added eosin Y, biological effects of eosin Y in a living body, and dyeing mechanism of cells.
