目的观察瓜蒌薤白半夏汤对心肌梗死大鼠Wnt信号通路中β-链蛋白(β-catenin)表达的影响。方法结扎大鼠左冠状动脉前降支建立大鼠心肌梗死模型,造模成功的大鼠随机分为心肌梗死模型组和瓜蒌薤白半夏汤组,每批次每组10只。心肌梗死模型组大鼠给予0.9%氯化钠注射液2 m L/次,每日1次灌胃,瓜蒌薤白半夏汤组大鼠给予浓煎中药2 m L/次,每日1次灌胃。分别干预1、5、10、15 d共4个批次。另取健康清洁级雄性SD大鼠10只,未行手术治疗,留为假手术组。免疫组化的方法检测β-catenin,IPWIN软件检测β-catenin表达率。结果心肌梗死模型组及瓜蒌薤白半夏汤组大鼠的β-catenin表达在心肌梗死后1、5、10、15 d均较假手术组升高,并且这2组大鼠在心肌梗死后分别干预5、10、15 d的β-catenin表达与假手术组比较差异均有统计学意义(P〈0.01)。与心肌梗死模型组大鼠比较,心肌梗死后给予瓜蒌薤白半夏汤干预1、5、10、15 d 4批次大鼠的β-catenin表达差异无统计学意义(P〉0.05)。结论大鼠的Wnt/β-catenin信号通路可被心肌梗死激活。短期使用瓜蒌薤白半夏汤未见到显著的促进心肌梗死大鼠心肌细胞β-catenin表达的作用。
Objective To explore the expression of beta- catenin after using Gualou- Xiebai- Banxia decoction in rats Wnt signaling pathway after myocardial infarction. Methods The left anterior descending coronary arteries of SD rats were ligated to establish myocardial infarction models. All rats with myocardial infarction were divided into model group( n = 10) and Gualou- Xiebai- Banxia decoction group( n = 10). The rats in control group were treated with 2 m L of 0. 9% Na Cl per day. The rats in Gualou- Xiebai- Banxia decoction group were treated with 2m L of concentrated decoction per day. There were four different duration of treatments specifically 1 d,5 d,10 d and 15 d. Another 10 healthy male SD rats were used as sham group. Beta- catenin was detected by immunohistochemical method. IPWIN software was used to detect the expression of beta- catenin. Results The expressions of beta- catenin 1 d,5 d,10 d and 15 d after myocardial infarction were increased in model and Gualou- Xiebai- Banxia decoction groups were higher than those in sham group,and the statistical differences existed in the durations of 5 d,10 d and 15 d after myocardial infarction( P〈0. 01). There were no statistical differences on the expressions of beta- catenin 1 d,5 d,10 d and 15 d after myocardial infarction between model and Gualou- Xiebai-Banxia decoction groups( P〉0. 05). Conclusion The Wnt / β- catenin signaling pathway in the rats may be activated by myocardial infarction. The short- term use of Gualou- Xiebai- Banxia decoction did not significantly increase the expression of beta- catenin in rats after myocardial infarction.
