从钝顶螺旋藻(Spirulina platensis)中提取纯化藻蓝蛋白(Phycocyanin,PC),光吸收值爿620朋280为3.1,观察Se(IV)与PC相互作用的光谱行为及红硒化现象。光谱检测发现,加入SeO3 2-后,Pc在620nm的特征光吸收减弱,并随Se(IV)浓度增加和时间延长单调降低,278nm和347nm的光吸收增强:PC的荧光发射和荧光激发也均呈现衰减。另外,PC与Na2SeO3相互作用后,观察到红硒化现象,透射电镜(TEM)证明主要存在形态为红色纳米硒。结果提示PC可能是SeO3 2-作用的敏感靶点.Se(W)对PC主要表现为氧化损伤,而Se(IV)被还原为红色纳米硒。
Spectra of interaction between Se(IV) and phycocyanin (PC), and red Selenium were explored in this paper. PC with an absorbance ratio of 3.1 (A620/A280) was purified from Spirulina platensis. The characteristic absorbance intensity of PC at 620 nm was decreased gradually after SeO3 2- was added, and the intensity decreased with the increases of the concentration and incubation time of Se (IV). But absorbance at 278 nm and 347 nm was gradually increased. As to fluorescence spectra of PC, both emission peak and excitation peak were decreased. Furthermore, the phenomenon of red selenium transformation was observed, and the dominant form of Se (0) was nano selenium as determined by TEM. These results suggested that PC might be a target of SeO3 2- Se (IV) had oxidative stress to PC and Se (IV) was reduced to red nano selenium.
