以高山红景天叶片为材料,研究了BA和NAA浓度对愈伤组织诱导的影响及MS培养基中氮和磷对愈伤组织增殖的影响。结果表明,在MS培养基中加入BA3mg/L和NAA0.3mg/L时高山红景天叶片愈伤组织的诱导效果好于其他BA和NAA浓度处理,诱导率达到62.2%。将诱导出的愈伤组织进行增殖培养时,将MS培养基中NH4+/NO3-配比由原有的20/40mM调节为30/30mM,PO43-浓度为1.25mM时,愈伤组织增殖生长良好,增殖系数达16.3。
The leaves of Rhodiola sachalinensis used as experimental material to study the effects of BA and NAA on callus induction as well as nitrogen and phosphorus concentration in MS culture medium on callus proliferation and growth. The results showed that the maximum callus induction was found in the medium supplemented with 3 mg/L BA and 0.3 mg/L NAA, the induction rate reached 62.2 %. The callus growth was affected by ratio of NH4 +/NO3- and phosphorus concentration,the favorable callus prolifera- tion and growth was examined when ratio of NH,+/NOa- in normal MS medium (20/40 mM) was adjus- ted to 30/30 mM. PO43- concentration in normal MS medium (1.25 mM) was suitable for callus prolifera- tion and growth, growth coefficient reached 16.3.
