为了研究促红细胞生成素产生肝细胞配体A1(Ephrin A1)对梅山猪胚胎附植期胚胎在子宫内膜间迁移和对子宫内膜黏附活动的影响,利用克隆测序、生物信息学和qRT-PCR方法,克隆了Ephrin A1基因的全长编码序列,对其表达的蛋白结构和功能进行了预测,并检测了其mRNA表达变化。梅山猪Ephrin A1基因编码区有5个c SNPs变异,其中2个导致氨基酸突变,分别为Glu29Asn和Trp33Arg,位于Ephrin A1外显子1和外显子2上。Ephrin A1编码序列中含5个外显子,编码205个氨基酸。Ephrin A1为不稳定的亲水性蛋白,有1个信号肽和一个保守区域,无跨膜区。另外,Ephrin A1在梅山猪胚胎附植期有较强的mRNA表达。表明Ephrin A1基因可能参与梅山猪的胚胎附植调控,其c SNPs有望成为影响猪产仔数基因的潜在分子标记。
In order to analyze the role of EphrinA1 in the migration and adhesion of embryo along the endometrium during embryo implantation in Meishan swine, EphrirtA1 coding sequence was cloned, protein structure and function were predicted and mRNA expression was detected by cloning, sequencing, bioinformatics and qRT-PCR method. Five cSNPs in EphrinA1 coding sequence were detected, two of which caused two amino acids changes which were Glu29Asn in exon 1 and Trp33Arg in exon 2. EphrinA1 coding se- quence contains five exons and encods 205 amino acids. Protein EphrinA1 is hydrophilic and instable, and has a signal peptide and a conservative region and no transmem- brane region. EphrinA1 mRNA showed a strong expression in Meishan swine during embryo implantation period. The results suggested that EphrinA1 might play a role in the regulation of embryo implantation in Meishan swine, and the cSNP is a potential molecular marker affecting swine litter size.
